It’s acceptable, but “please do two things for me” is standard. In English an indirect object can be implied by placing the object (pro)noun between the verb and the direct object, as in “I baked Jane a cake” or “he did me a favor last week”, which are equivalent to “I baked a cake for Jane” and “he did a favor for me last week”.

If you are going so far as to say “would you please”, you are probably better off with the more formal construction.

If you say “would you please do me” it means would you please have sex with me. (Do me = Screw me or have sex with me) usually the woman will say “do me” and the guy says “I’m going to do you” But a guy can say “do me” in “safe” situations where it can’t be heard by another man in which case it might sound like the first man is saying to the the second man “do me in the butt.” And unless you like that sort of thing it’s generally frowned upon by straight men.

Then if you add …”two things” it doesn’t really make sense and it could be heard as “do me WITH two things” and then you’re really in trouble (again, unless you like that sort of thing… er, things)

HOWEVER very few people speak perfect English, so you can probably get away with saying “Would you please do me two things” and people will know what you mean. But you won’t impress your English Professor.

Though you might try doing HIM with two things. You never know, he might like that sort of thing.

No. It is not acceptable. Beckers is correct.

You could say “Would you please do me two favours?” or “Would you please do me two services?” but these are rather special cases. Doing someone a favour, or doing someone a service are acceptable. Doing a thing is not.
