Can the word ‘comprise’ be used with the preposition ‘of’?
‘comprise of’ is often used in conversation.Is it correct or should only ‘comprise’ be used?

No, it can’t. Unless of course you’re using it in the passive.
Examples: (active voice) The class comprises 25 students (the class consists of 25 students). You can’t say “the class comprises of 25 students.”
However, in the passive, you can say: the class is comprised of 25 students (uncommon, but correct)

Neel is right. When you use “comprise” in the passive, you use “is [or was] comprised of.” In the active, you would not use “of”.
