Although “The Crucible” has many themes, the theme of hypocrisy is the most important one.
(Don’t use the word SUBJECT when you mean THEME.)

You don’t need any extra punctuation in there. Putting something in there will only add unnecessary bulk to the sentence. If you’re ever in doubt, just read the sentence out loud. Most people wouldn’t put a pause before “hypocrisy.”

well it depends on if you are just writting about hypocrisy or if you want to include other “subjects”. you need to decide which on b/c it will depend on if you just want to write about the hypocrisy or not.
